NanoString GeoMx® Digital Spatial Profiler (DSP)

The GeoMx® Digital Spatial Profiler (DSP) by NanoString is a cutting-edge tool designed for high-plex spatial and molecular analysis of RNA and protein within tissue samples. This innovative platform empowers researchers to connect spatial context with detailed molecular data, offering new insights into tissue biology.

Technology Overview:

  • Spatially Resolved Profiling: The GeoMx DSP employs panels of UV-cleavable DNA barcoded probes to target RNA or protein in tissue sections. These probes are combined with fluorescently labeled morphology markers, which illuminate the structural features of the tissue, enabling precise region of interest (ROI) selection based on visual guidance.
  • Targeted Data Collection: After selecting ROIs, UV light is used to release the DNA barcodes from the targeted areas. These barcodes are then collected and quantified using NanoString's nCounter system, providing a detailed molecular profile specific to the selected ROI. For broader profiling needs, GeoMx can also be paired with next-generation sequencing (NGS) to analyze over 18,000 targets on a single slide.

Key Features:

  • Multi-Omic: Analyze RNA and protein with any sample types, such as formalin-fixed paraffin-embedded (FFPE), fresh-frozen tissue sections, core needle biopsy, Slide mounted tissue, cell lines, and tissue microarray, making it adaptable to various research needs.
  • Whole-Slide Imaging: Allows for comprehensive imaging with up to four fluorescent stains to accurately guide the selection of ROIs.
  • Wide Dynamic Range: The platform supports up to 6 logs (base 10) of dynamic range, ensuring precise digital quantitation.
  • Preservation of Samples: The non-destructive processing preserves tissue integrity for potential further analysis.
  • Broad Applicability: Ideal for exploring complex biological questions in fields such as immunology, oncology, and neuroscience, with continuously expanding content options, including the GeoMx Whole Transcriptome Atlas.
